Multi-part question: 1) How many missiles are required to kill each boss? 2) To have that many missiles (or close to that many), which expansions are the quickest, and the most along-the-way?
I'll answer the first part of your question. Kraid takes 15 missiles(or 3 super missiles). Ridley takes 50 missiles(or 10 super missiles). Mother Brain takes 55 missiles, 20 for the glass and 35 for the eye(or a total of 11 super missiles). Mecha Ridley takes 30 missiles, 10 for the glass shield and 20 to destroy the body(or 6 super missiles) while it's 90 missiles in total for a 100% run with 30 for glass shield and 60 to destroy body(or 18 super missiles).
Depends on your definition of quick-kill. Technically, he isn't fully risen by the time you get 3 supers in his mouth.
Also, in Kraid's case, it takes more like 18 or 19 missiles (17 if you're really good), because his eye doesn't take damage, it just opens his mouth.
In Normal mode at least (keep in mind I'm a huge promoter of Kraid-first), I personally take the first missile tank, the one in the hive room, the one in the hall after that that you basically run right through normally, grab the etank as I return from bombs, and then go after Kraid. Considering you have to shoot his eye to open his mouth regardless, I just get missile refills from his fingernails as I fight him, so 15 is enough capacity.
From there I get the tank immediately after the elevator, the etank in skree-hall for insurance (just for Ridley), the missile in the secret tunnel in the middle of the brinstar shafts, the one after the...gravity suit I think it is?, shinespark for the super immediately after the grav suit, the missile in the hall returning from grip since it's fast just gripping up there, the one in the hall above ice cream right before the Norfair secret shaft, the one at the bottom of said secret shaft (that AJ inexplicably skips in his hard mode run O.o), the one in the fireball hall, the super right above the same hall, the missile dividing the 2 supers, the super on the other side of said missile, the etank behind space jump (?) before refilling my ammo at the chozo statue, the etank in the hall directly east of Ridley for insurance because if I'm doing normal I'm probably streaming live, then I shinespark up the Norfair shaft and leave.
My capacity is 55/6 at this point. I also get the super missile immediately after the Chozo Test for insurance against Iron Ted, but if I don't need them on him, I use them on the drone at the start of the escape spark preparation.
I'm still deciding for Hard mode, but I skipped 2 aj got and got 2 aj didn't. I might look for more. ;>_>

True, but nobody ever call it the 'quick kill' in ZM. I have only used supers a couple of times against Kraid in ZM, because it requires a difficult and/or time consuming route to get the supers before Kraid. In Super, you're kinda supposed to have super missiles at the Kraid fight.
Tyjet, it's not necessarily beneficial, it's just that the TASes have Ridley first as being 10 seconds faster (I think? I could be mistaken), and uses the grip a ton throughout Kraid in a manner no human can even come close to replicating. Due to this, I propose that Kraid first is roughly as fast as Ridley first if performed on console, and yet almost nobody ever does Kraid first runs.
(and yeah I'm talking about any% normal)
EDIT: Okay maybe not a ton. The biggest use of it is to perform the escape spark and not need to go to the hall beneath the rio room to charge said spark, due to the speed of climbing up the crumble blocks.

If you do that trick in the rightmost room in norfair to fall down the shaft, you never go through a heated area.

I don't get varia until after kraid.
I never even grab it.
I do since it's right there anyways. I figure it only adds like 5 seconds to grab it, and it does come in handy.
Varia? Doesn't it add a couple minutes? Back in the days I used to get it, I IBJed up the shaft immediately after bombs and ohhhh you're talking about 100%. I see. In 100%, is the acid dive done instead of DBJing for the Varfia? It probably adds more like 15 seconds, though.
By the time I get varia, I have power grip and hi-jump/sringball so I easily take the high road across the room to the varia, then dive into the acid to grab the ET on the way back. I'm not sure about how many seconds it takes to run over into the room with the varia in it, but I'm going so close to it anyways since I'm getting the ET that I think the time is pretty small. I realize that I don't need to grab the varia in order to get 100%, but to be honest I don't think I have tried it without getting it. Not sure what kind of crap I would have to change about my current route to avoid heated areas and such.
